CDKs play important roles in the control of cell division and modulate transcription in response to several extra- and intracellular cues. Additionally, the LxF motif is critical for targeting M-CDK to phosphorylate several mitotic regulators; for example, Spo12 is targeted via LxF to release the phosphatase Cdc14. The results complete the full set of G1, S, and M-CDK docking mechanisms and outline the unified role of cyclin specificity and CDK activity thresholds. The Cdk-cyclin complexes regulate a series of events that lead cells from a resting state (G0), growth phase (G1), through DNA replication (S), and finally to cell division (M). Abnormalities in cell cycle control that occur in any of the phases leads to cell cycle arrest and might be associated with cancer 1 . Mitotic-cyclin (M-cyclin) levels steadily increase in the G2 phase and peak right before mitosis, at which point M-Cdk is activated, driving the cell into the M phase (Figure 6).

Systems Biology of the Cell Cycle A “Baby” of UNICELLSYS
The mammalian cell cycle is driven by a complex of cyclins and their associated cyclin-dependent kinases (CDKs). Abnormal dysregulation of cyclin-CDK is a hallmark of cancer.

A cyclin-dependent kinase (CDK) binds a regulatory protein called a Cyclin. Without cyclin, CDK has little kinase activity; only the cyclin-CDK complex is an Cyclin-CDK complexes are activated by phosphorylation of the CDK subunit by cyclin-dependent activating kinase (CAK) that contains three subunits: CDK7, cyclin H, and MAT1. By contrast, cyclin-CDK complexes are inhibited by phosphorylation in adjacent threonine and tyrosine residues by the dual-specificity protein kinases, WEE1 and MYT1. The eukaryotic cell cycle is regulated by the temporal activation of different cyclin-dependent kinase (CDK)/cyclin complexes. Whilst the level of the catalytic subunit of the complex, the CDK, remains relatively constant through the cycle, the level of the cyclin subunit generally oscillates.

Cyclin-dependent kinase, or CDK, is a type of enzymatic protein that resides in eukaryotic cells and plays a key role in cellular metabolism and renewal, a series of biological processes collectively referred to as the cell cycle. Several protein kinases are also classified as genes, making up about 2 percent of all human genes. Cell changes in the cell cycle like the assembly of mitotic spindles and alignment of sister-chromatids along the spindles are induced by M cyclin- Cdk complexes.
This interaction leads to phospho-adaptor Cks1-mediated inhibition of M-CDK to facilitate Cdc6 accumulation and sequestration in mitosis. The LxF motif and Cks1 also mediate the mutual inhibition between M-CDK and the tyrosine kinase Swe1.
